5a/6a Promoter Gene Polymorphism and Risk of Myocardial Infarction in South Indian Population

چکیده
Introduction: Myocardial infarction (MI) is a multifactorial disease influenced by environmental and genetic factors. Matrix metallo proteinase-3 (MMPs) plays a pivotal role in the development of atherosclerosis and MI. Objective: The objective of the present study was designed to investigate the association of matrix metallo proteinase3 -1612 promoter 5A/6A gene polymorphism. Methods: In the present study was carried out with 250 myocardial infarction patients and 260 controls. Clinical and demographic characteristics were collected.DNA was isolated and MMP-3 -1612 promoter 5A/6A gene polymorphism was investigated using polymerization reaction followed by restriction digestion for all samples. Results: The percentage of classical risk factors like body mass index, hypertension, smoking, alcohol and diabetes was high in patients when compared to controls. The matrix metallo proteinase 3 -1612 promoter 5A gene polymorphism was not associated with myocardial infarction. Conclusion: The MMP-3 promoter 5A/6A gene polymorphism is not a risk factor for myocardial infarction patients in a SouthIndian population

Stromelysin promoter 5A/6A polymorphism is associated with acute myocardial infarction.
BACKGROUND Rupture of the fibrous cap of an atherosclerotic plaque is a key event that predisposes to acute myocardial infarction (AMI). Matrix metalloproteinases (MMPs) may contribute to weakening of the cap, which favors rupture. Stromelysin, a member of MMP family, is identified extensively in human coronary atherosclerotic lesions. It can degrade most of the constituents of extracellular ma...
متن کاملMMP3 -1171 5A/6A Promoter Polymorphism Affects Level of Serum Major Histocompatibility Complex Class Chain Related B in Breast Cancer Patients
Background and Aims: Involvement of matrix metalloproteinases 3 (MMP3) in breast cancer tumor progression and metastasis has been revealed. MMP3 -1171 5A/6A and 5A/5A polymorphisms in the gene promoter increase expression of the enzyme. The possible relationship of these polymorphisms and serum levels of major histocompatibility complex class I chain-related protein A and B (MICA/B) in br...

Objective: To study the associations of stromelysin-1 (MMP3) gene 5A/6A polymorphism with plasma high-sensitivity C-reactive protein (hs-CRP) level in coronary artery disease (CAD) and myocardial infarction (MI). Methods: The MMP3 5A/6A genotypes and plasma hs-CRP levels were determined in 405 non-CAD subjects and 395 angiography-documented CAD patients, 157 with MI and 238 with non-MI. Results...
متن کاملThe Stromelysin-1 5A/6A Promoter Polymorphism Is a Disease Marker for the Extent of Coronary Heart Disease
BACKGROUND Matrix metalloproteinases, such as stromelysin-1, are implicated in the pathogenesis of coronary artery disease (CAD) and acute myocardial infarction (MI). A 5A/6A promoter polymorphism can regulate the transcription of the stromelysin-1 gene in an allele-specific manner. Evidence has been presented that the 6A allele is associated with the progression of coronary heart disease (CHD)...
متن کاملThe Role of Matrix Metalloproteinase-3 Functional 5A/6A Promoter Polymorphism in Tumor Cell Progression and Metastasis of Breast Cancer
In the human genome, chromosome 11 contains a cluster of matrix metalloproteinase (MMP) genes. Single nucleotide polymorphisms in the promoter region of MMP genes are important for MMP expression. A common adenine deletion polymorphism (5A) at position -1171 of the MMP-3 gene promoter (5´-AAAAAACCAT-3´ change to 5´-AAAAACCAT-3´) facilitates transcriptional factor binding and MMP-3 promoter acti...
